Effects of c-FLIPL Knockdown in Cervical Uterine Carcinoma Cell Lines International Journal of Morphology
Ili,Carmen G; Brebi,Priscilla; Garcia,Patricia; Leal,Pamela; Lopez,Jaime; Tapia,Oscar; Letelier,Pablo; Weber,Helga; Castillo,Jonathan; Roa,Juan C.
Overexpression of Short and Raji variants of Cellular FLICE-like inhibitory protein (c-FLIP) is capable of inhibiting apoptosis, while the function of the Long isoform depends of c-FLIPL concentration in cells. The aim of this study was to determine the effects of c-FLIPL knockdown in cervical cell lines. SiHa, C-4I and C-33A cervical cancer cell lines were analyzed. c-FLIPL level expression was determined by quantitative real-time PCR and western blotting. c-FLIPL was transiently downregulated by siRNA. The effects of knockdown of c-FLIPL on cell viability, proliferation and apoptosis were assessed by comparing with scrambled siRNA-transfected cells. SiHa and C-4I c-FLIPL knockdown cells showed increased viability compared with scrambled siRNA-transfected...

Effects of Epstein-Barr virus on the development of dendritic cells derived from cord blood monocytes: an essential role for apoptosis BJID
Wang,Juan-Juan; Li,Yu-Feng; Jin,Ying-Ying; Wang,Xi; Chen,Tong-Xin.
OBJECTIVE: Epstein-Barr virus (EBV) is a ubiquitous human γ-herpes virus, which can adapt and evade host immune defense. Dendritic cells (DCs) play a pivotal role in the initiation and maintenance of immune responses. This study investigated the effects of EBV on cord blood monocytes derived DCs (CBDC). METHODS: Monocytes were isolated from cord blood and cultured in medium containing recombinant IL-4 and GM-CSF to induce DCs development. B95-8 supernatant was added in monocytes culture medium for EBV infection at day 0. Phenotypic characterization of DCs, apoptotic cells, and mitochondrial membrane potential (MMP) were detected by flow cytometry. The morphology was observed by Hoechst 33258 staining and TUNEL staining, the expression of X-linked inhibitor...

Effects of karanjin on cell cycle arrest and apoptosis in human A549, HepG2 and HL-60 cancer cells Biol. Res.
Guo,Jian-Ru; Chen,Qian-Qian; Lam,Christopher Wai-Kei; Zhang,Wei.
BACKGROUND: We have investigated the potential anticancer effects of karanjin, a principal furanoflavonol constituent of the Chinese medicine Fordia cauliflora, using cytotoxic assay, cell cycle arrest, and induction of apoptosis in three human cancer cell lines (A549, HepG2 and HL-60 cells). RESULTS: MTT cytotoxic assay showed that karanjin could inhibit the proliferation and viability of all three cancer cells. The induction of cell cycle arrest was observed via a PI (propidium iodide)/RNase Staining Buffer detection kit and analyzed by flow cytometry: karanjin could dose-dependently induce cell cycle arrest at G2/M phase in the three cell lines. Cell apoptosis was assessed by Annexin V-FITC/PI staining: all three cancer cells treated with karanjin...

Effects of propofol on damage of rat intestinal epithelial cells induced by heat stress and lipopolysaccharides BJMBR
Tang,J.; Jiang,Y.; Tang,Y.; Chen,B.; Sun,X.; Su,L.; Liu,Z..
Gut-derived endotoxin and pathogenic bacteria have been proposed as important causative factors of morbidity and death during heat stroke. However, it is still unclear what kind of damage is induced by heat stress. In this study, the rat intestinal epithelial cell line (IEC-6) was treated with heat stress or a combination of heat stress and lipopolysaccharide (LPS). In addition, propofol, which plays an important role in anti-inflammation and organ protection, was applied to study its effects on cellular viability and apoptosis. Heat stress, LPS, or heat stress combined with LPS stimulation can all cause intestinal epithelial cell damage, including early apoptosis and subsequent necrosis. However, propofol can alleviate injuries caused by heat stress, LPS,...

Effects of silk sericin on the proliferation and apoptosis of colon cancer cells Biol. Res.
Kaewkorn,Waraporn; Limpeanchob,Nanteetip; Tiyaboonchai,Waree; Pongcharoen,Sutatip; Sutheerawattananonda,Manote.
Sericin is a silk protein woven from silkworm cocoons (Bombyx mori). In animal model, sericin has been reported to have anti-tumoral action against colon cancer. The mechanisms underlying the activity of sericin against cancer cells are not fully understood. The present study investigated the effects of sericin on human colorectal cancer SW480 cells compared to normal colonic mucosal FHC cells. Since the size of the sericin protein may be important for its activity, two ranges of molecular weight were tested. Sericin was found to decrease SW480 and FHC cell viability. The small sericin had higher anti-proliferative effects than that of the large sericin in both cell types. Increased apoptosis of SW480 cells is associated with increased caspase-3 activity...

Zhang,Y; Yang,H; Wu,X; Deng,M; Li,Z; Xu,Z.
ABSTRACT Avian leukosis virus subgroup J (ALV-J), a member of the retroviridae family, can infect both broilers and layers and induce a spectrum of different neoplasms, resulting in serious economic losses in poultry production. Epigallocatechin-3-gallate (EGCG), the major constituent of green tea, has demonstrated remarkable anti-inflammatory and cancer chemopreventive effects in many animal tumor bioassays, cell culture systems and epidemiological studies. To assess the antiviral effects of EGCG on ALV-J-induced cell apoptosis in vitro, DF-1 cells were treated with different EGCG concentrations (0, 5, 10, 20 and 40 µg/mL), and their antiviral effects were examined at different time points (0, 24, 48, 72 and 96 h) using a variety of assays. EGCG...

Lv,Dan; Pan,Li-hong; Zhang,Ren; Yang,Jie; Chen,Hao; Wen,Yanzhang; Huang,Mi; Ma,Xinhua; Wang,Qiang; Yang,Xinzhou.
Hepatocellular carcinoma is one of the most prevalent malignancies and a leading cause of cancer-related mortality worldwide. However, the therapies to prevent hepatocellular carcinoma are still limited and the emergence of drug resistance leads to the development of new anti-cancer drugs and combinational chemotherapy regimens. Our study was aimed to explore the anticancer effects of the essential oil extract (EEEO) from Euphorbia esula which has been widely used in traditional Chinese folk medicine and possessed potential cytotoxic effects in several human tumor cells. However, the mechanisms of EEEO-induced anti-proliferation and apoptosis have not been completely elucidated. In this study, EEEO was prepared by hydro-distillation and the main chemical...

Gervais, Ophelie.
The flat oyster, Ostrea edulis, is the European endemic oyster species. Its production has been reduced because of overfishing and diseases including bonamiosis. Massive mortalities observed on the Pacific cupped oyster these last years explained the wish of shellfish farmers to diversify their production and their revival of interest for this patrimonial oyster species. However, bonamiosis due to the protozoan, Bonamia ostreae, is still a major problem for the production of this species. Measures to control the disease are limited and require a better knowledge of the interactions between the flat oyster and the parasite. Previous studies have suggested the involvement of apoptosis in flat oyster defense mechanisms against B. ostreae. This mechanism is...

Etude de nouvelles cibles moleculaires de cancer bronchopulmonaire non a petites cellules pharmacomodulées par des substances originales naturelles et synthetiques ArchiMer
Moreau, Dimitri.
Still rare disease at the beginning of the 20th century, the lung cancer is at the origin of more than 6 % of the deaths in France currently. It occupies there the first rank of the deaths by cancer whereas France is regarded as a country at the average risk among the Western countries. The discovery of new anti-cancer treatments increasingly more effective and specific, is thus today of primary importance. The validation of the capacity of a molecule to being able to inhibit the cancerous proliferation, passes by several stages of tests starting with the description of an activity at the cellular level. One of the strategies used for the discovery of new compounds antiproliferativs, passes by an investigation of the bioactiv potential of a great number...

Evaluation of annexin V and Calcein-AM as markers of mononuclear cell apoptosis during human immunodeficiency virus infection BJID
Palma,Priscila F.R.; Baggio,Giovana L.; Spada,Celso; Silva,Renata da; Ferreira,Silvia Inês A.C.P.; Treitinger,Aricio.
Evaluation of apoptosis by flow cytometry is generally accomplished by methods that use annexin V-FITC as vital dye, which access phosphatidylserine exposed on the external membrane at the beginning of this process. In addition, the concomitant use of propidium iodide makes possible to verify the characteristic nuclear alterations in the late stages of apoptosis, as a consequence of the increase in membrane permeability. On the other hand, the use of calcein-AM in association with ethidium homodimer (EthD-1) allows the evaluation of cell apoptosis through detection of esterase activity and cellular membrane physical and chemical alterations. The aim of this study was to compare the sensibility of calcein-AM and EthD-1 with annexin V-FITC and propidium...

Rudge,Marilza Vieira Cunha; Costa,Elaine; Barbisan,Luis Fernando; Damasceno,Débora Cristina; Bueno,Aline; Saito,Felipe Hiroshi; Calderon,Iracema Mattos Paranhos; Rodrigues,Marcela Marcondes Pinto.
The aim of this work was to analyze the cell proliferation and apoptosis indexes on the 18th and 21st days of pregnancy of diabetic rats and to correlate with maternal glycemia and perinatal outcomes. Placentas from 20 Wistar rats were collected and divided into four experimental groups: control and diabetic of 18 and 21 days of pregnancy. The cell proliferation was analyzed using the PCNA expression and apoptosis by the TUNEL method. It was observed that PCNA and TUNEL indexes decreased from day 18 to 21 of pregnancy in the placentas of diabetic rats and these values were lower than control groups. Diabetic dams presented higher percentage of small for pregnancy age (SPA) fetuses. However, there was no difference between the PCNA and TUNEL indexes in SPA...

Eighty-six newly diagnosed multiple myeloma (MM) patients from a public hospital of São Paulo (Brazil) were evaluated by cIg-FISH for the presence of del(13)(q14), t(4;14)(p16.3;q32) and del(17)(p13). These abnormalities were observed in 46.5, 9.3, and 7.0% of the patients, respectively. In order to identify the possible role of del(13)(q14) in the physiopathology of MM, we investigated the association between this abnormality and the proliferative and apoptotic indexes of plasma cells. When cases demonstrating t(4;14)(p16.3;q32) and del(17)(p13) were excluded from the analysis, we observed a trend towards a positive correlation between the proportion of cells carrying del(13)(q14) and plasma cell proliferation, determined by Ki-67 expression (r = 0.23, P...

Evasion of immune responses by Trypanosoma cruzi, the etiological agent of Chagas disease BJMBR
DosReis,G.A..
Infection with the protozoan parasite Trypanosoma cruzi leads to Chagas disease, which affects millions of people in Latin America. Infection with T. cruzi cannot be eliminated by the immune system. A better understanding of immune evasion mechanisms is required in order to develop more effective vaccines. During the acute phase, parasites replicate extensively and release immunomodulatory molecules that delay parasite-specific responses mediated by T cells. This immune evasion allows the parasite to spread in the host. In the chronic phase, parasite evasion relies on its replication strategy of hijacking the TGF-β signaling pathway involved in inflammation and tissue regeneration. In this article, the mechanisms of immune evasion described for T. cruzi...

Excretion Product of Shigella dysenteriae (SdyEP) Induced Cell Death in Early Larval Stage of Zebrafish (Danio rerio): Acridine Orange and Ethidium Bromide (AO/EB) in vivo Staining International Journal of Morphology
Alvarez,M; Urbina,G; Perdomo,L.
In the field of studies of acute toxicity induced by bacterial agents, Shiga toxins have been relevant due to the severity of the extra-intestinal diseases they cause. Numerous studies have shown that Shiga toxin induced apoptosis in different cell types; however, this important process has been little studied in vivo experimental models. In this study, the effects of excretion products of Shigella dysenteriae, in which Shiga toxin is present, were investigated on early larval stages of Zebrafish, an animal model with many advantages over other in vivo experimental models traditionally used. Both the collection of eggs and larvae of Zebrafish, and the product from excretion from Shigella dysenteriae (SdyEP) were performed according to laboratory standards....
